How it works

Inhibits focal adhesion kinase, reducing stromal fibrosis and remodeling. Hypothesized to improve gemcitabine delivery in mPDAC. Under investigation in ACCENT trial.

Investigational FAK inhibitor. RP2D is 400 mg PO once daily with food in a 28-day cycle alongside D1/D8/D15 GnP. The reported DLT in the 400 mg cohort was Grade 3 nausea. No prompt-engine claim is made about narmafotinib-specific LFT monitoring beyond the standard GnP backbone.

Dosing

Typical doses

  • 400 mg PO once daily (RP2D, ACCENT trial)

Schedule

  • 400 mg once daily with food, continuous throughout 28-day cycle
    with_meals

Side Effects

  • Nausea, vomiting
  • Fatigue
  • Diarrhea or loose stools
  • Grade 3 nausea was the dose-limiting toxicity reported at 400 mg in the ACCENT phase 1b cohort

Monitoring

  • Adherence to once-daily dosing with food — investigational oral agent
  • Standard chemotherapy laboratory monitoring per the GnP backbone (FBC, LFTs, U&Es D1/D8/D15)
